Design Rules for Vertical Paper-based Immuno-Diagnostic System (VPI-DS)

A systematic approach, including quality risk management (QRM) and design of experiments (DOE), is used to reduce signal variation across the paper membrane and to evaluate CPPs on sensitivity of VPI. Biomarkers for five Tier 1 bacterial pathogens will be used as model systems for the study. The cAbs arrays will be printed, biomarker purified, mAbs purified and affinity/dissociation constant characterized to support the project. LFIs will be developed to compare with the corresponding VPIs. A theoretical paper-based biosensor model is proposed and will be validated. Design rules will be generated for both single-plex and multiplex VPIs. Sponsor: DoD/DTRA.
