  • Time-resolved interrogation of organellar proteostasis under unfolded protein response

Disrupted proteostasis is a hallmark of various human diseases including cancer, heart disease and neurodegeneration. Different cellular compartments have evolved specialized machinery to handle specific stressors, including the endoplasmic reticulum unfolded protein response (UPR) that is triggered by ER stress; however, the full scope of these responses remains unclear. Recent evidence suggests that there exists a complex temporal hierarchy of proteomic changes that extend far beyond the initial activation of UPR pathways. In this presentation, I will describe our use of subcellular spatiotemporal proteomics to distinguish between the abundance of pre-existing and newly synthesized proteins across multiple compartments. I will highlight a recently optimized method, SPLAT-TR (Simultaneous Proteome Localization and Turnover analysis with Time Resolution), which enables multidimensional measurements of protein abundance, synthesis, degradation, and localization during the time-evolution post-UPR. We will show evidence that ER protein misfolding propagates stress to multiple organelles, triggering distinct changes in protein quality control.
